Diagnosis

The quantitative buffy coat technique (QBC® tube) is a method that uses fluorescent dyes in a capillary tube that is centrifuged and examined by ultra-violet/fluorescence microscopy (top). With this technique, the nucleus of parasites infecting red blood cells fluoresces bright green and while its cytoplasm is yellow-orange (bottom). Other tests include enzyme-linked immunosorbent assay, radioimmunoassay, rapid immunochromatographic dipstick tests, tests for P. falciparum histidine-rich protein-2 and tests for parasite lactate dehydrogenase. Molecular techniques include DNA hybridization and polymerase chain reaction methods. Indirect fluorescent tests for detection of antimalarial antibodies in serum cannot distinguish current and prior infections and are used primarily in epidemiological studies.
